Abstract:
In this study, a dual-recombination system allowing sequential manipulation of KrasG12D-driven pancreatic cancer (PDAC) was characterized. For secondary manipulation, the Flp/frt recombination system was combined with an inducible Cre/loxP system. The efficiency of this model was proven by reconstitution of p53 in tumor cell lines. In a second step, the tumor microenvironment was deciphered using the dual-recombination system. It could be shown that mast cells are dispensable for PDAC development and progression.

In dieser Arbeit wurde ein duales Rekombinationsmodell charakterisiert, mit welchem eine sequentielle Manipulation im KrasG12D-abhängigen Pankreaskarzinom (PDAC) möglich ist. Das Flp/frt Rekombinationssystem wurde mit dem induzierbaren Cre/loxP kombiniert. Die Effektivität des neuen Modells konnte mittels Rekonstitution des p53 Proteins in Tumorzelllinien gezeigt werden. Im zweiten Teil der Arbeit wurde das Modell verwendet um das Tumormikromileu zu charakterisieren. Es wurde gezeigt, dass die PDAC-Entstehung und Aufrechterhaltung des Tumors unabhängig von Mastzellen ist.
