This is the first study precisely determining the absorbed doses in the peaks and valleys of microbeams using the CBMN-Assay. The biological dosimetry with micronuclei and bridges agrees with the physical dose determined with Gafchrom film and could therefore validate physical dosimetry. The study clearly demonstrated that reconstructed skin tissue is well suited to study spatial resolution of DNA damage after microbeam therapy. Here, a higher 53BP1 foci number could be observed in the valleys compared to homogeneous irradiation, which could indicate possible bystander effects.
