Kurzfassung:
Nanog is a key regulator of pluripotency in mouse embryonic stem cells (mESCs). In this thesis I present analysis of Nanog dynamics including transitions between fluorescence intensity compartments, oscillations, and onsets of Nanog expression. I present evidence for mESC subpopulations differing in expression dynamics and pluripotency factor correlation structures. I then develop mechanistic models for onset dynamics and a tree-based Bayesian inference algorithm for stochastic autoregulatory gene expression models.
