Kurzfassung:
By tethering proteins to a surface via dynamically actuated DNA nanolevers and comparing their motional dynamics to a theoretical model, protein diameters can be quantified with Angstrom accuracy. Alterations in the tertiary protein structure and conformational changes are revealed and the applicability for the affinity ranking of small molecules and the characterization of DNA polymerases are demonstrated. The presented method eliminates the need to run multiple protein assays sequentially and might facilitate improved workflows in drug discovery and protein engineering.

Veränderungen in Tertiärstruktur und Konformation werden aufgedeckt und ein Affinitätsscreening von kleinen Molekülen sowie die Charakterisierung von DNA Polymerasen wird demonstriert.
