Abstract:
Compromised function of the mRNA translation machinery often leads to tissue-specific diseases. To understand the underlying mechanisms, we queried the importance of its components in proliferating and maturated human cells with CRISPRi screens. The loss of translation-dependent quality control factors leads to phenotypes that are strongly context-dependent. Furthermore, we identified new endogenous human mRNAs that are substrates of co-translational quality control pathways.

Eine gestörte Funktion der mRNA-Translationsmaschinerie führt häufig zu gewebespezifischen Krankheiten. Um die zugrundeliegenden Mechanismen zu verstehen, haben wir ihrer Komponenten in proliferierenden und adulten menschlichen Zellen mithilfe CRISPRi-Screens untersucht. Der Verlust von Qualitätskontrollfaktoren führt zu Phänotypen, die stark kontextabhängig sind. Darüber hinaus haben wir neue endogene mRNAs identifiziert, die Substrate dieser co-translationalen Qualitätskontrolle sind.
