Kurzfassung:
The spread of antibiotic-resistant Staphylococcus aureus urgently demands the development of novel antibiotics. Structure-activity relationship studies of the human kinase inhibitor sorafenib led to the discovery of a new potent antibiotic compound, PK150. The therapeutic potential of PK150 was studied by in vitro and in vivo experiments, revealing a promising pharmacological profile. Additionally, target identification by chemical proteomics and further studies regarding the antibiotic mode of action were conducted.

Struktur-Aktivitätsbeziehungsstudien des humanen Kinaseinhibitors Sorafenib führten zur Entdeckung einer neuen potenten antibiotischen Verbindung, PK150. Das therapeutische Potenzial von PK150 wurde durch In-vitro- und In-vivo-Experimente untersucht und zeigte ein vielversprechendes pharmakologisches Profil. Zusätzlich wurden die zellulären Angriffsziele der Verbindungen durch chemische Proteomik analysiert und weitere Studien zur Aufklärung der antibiotischen Wirkungsweise durchgeführt.
