Abstract:
In einem multimethodischen Ansatz wurden spezifische molekulare Einflussfaktoren auf die Allergenität des Birkenpollen-assoziierten Apfelallergens Mal d 1 untersucht. Dazu wurde der Mal d 1 Gehalt in Apfel-Genotypen verschiedener Kreuzungspopulationen mittels indirekt kompetitiven ELISA quantifiziert. Proteomics ermöglichte die Identifizierung von Peptid-/Isoformmarkern, die möglicherweise hypo- und hyperallergene Eigenschaften besitzen. Daneben wurden Bindungspartner von Mal d 1 mit Bindungsaffinitäten im niedrigen mikromolaren Bereich identifiziert.

In a multi-methodological approach, specific molecular factors influencing the allergenicity of the birch pollen-associated apple allergen Mal d 1 were investigated. The Mal d 1 content was determined in apple genotypes of different crossbreeding populations using an indirect competitive ELISA. Proteomics revealed peptide/isoform markers that may be affiliated with hypo- and hyperallergenic properties. Moreover, ligands of Mal d 1 with binding affinities in the low micromolar range were identified.
