In this work, the mechanism of action of the natural product xanthocillin X was elucidated, which exhibits impressive activity against numerous bacterial pathogens. As it turned out, xanthocillin X binds to the cofactor heme and a dysregulation of the heme biosynthesis is the lethal consequence.
In the second part of the work, probes based on the pyridoxal scaffold have been developed, which allow a reliable detection of pyridoxal kinases in bacteria that form a hemithioacetal with pyridoxal during catalysis.

Im Rahmen dieser Arbeit wurde der Wirkungsmechanismus des Naturstoffs Xanthocillin X aufgeklärt, der eine beeindruckende Aktivität gegen zahlreiche bakterielle Krankheitserreger aufweist. Wie sich herausstellte, bindet Xanthocillin X an den Cofaktor Häm und eine Dysregulierung der Häm-Biosynthese ist die letale Folge.
Im zweiten Teil der Arbeit wurden Sonden basierend auf dem Grundgerüst des Pyridoxals entwickelt, mit denen Pyridoxalkinasen, die bei der Katalyse mit Pyridoxal ein Hemithioacetal bilden, in Bakterien zuverlässig nachgewiesen werden können.
