Abstract:
Within this thesis, it was feasible to synthesize a new functional initiator for rare earth metal-mediated group transfer polymerization of dialkyl vinylphosphonates. This functional group enabled the end-group functionalization of the polymers with biomolecules via thiol-ene click chemistry. The folic acid and cholesterol conjugates showed the retention of the biological properties of the linked biomolecules. In the second part of the thesis, novel cross-linked polyvinylphosphonate nanoparticles were synthesized successfully. The particles are spherical, monodisperse and in the desired size range for biomedical applications. Their loading and release properties, as well as their biocompatibility showed promising results. In subsequent in vitro studies, the delivery of a chemotherapeutic agent into cancer cells was achieved.

Im Rahmen dieser Arbeit war es möglich, einen neuen, funktionalen Initiator für die seltenerd-mediierte Gruppentransferpolymerisation von Dialkylvinylphosphonaten zu synthetisieren. Die funktionelle Gruppe ermöglichte die Endgruppenfunktionalisierung der Polymere mit Biomolekülen durch die Anwendung der Thiol-En-Klick-Chemie. Die Folsäure- und Cholesterolkonjugate zeigten die Erhaltung der biologischen Eigenschaften der gebundenen Biomoleküle. Im zweiten Teil der Arbeit wurden neuartige, quervernetzte Polyvinylphosphonat-Nanopartikel synthetisiert. Die Partikel sind sphärisch, monodispers und besitzen die vorausgesetzte Größe für biomedizinische Anwendungen. Ihre Aufnahme- und Freisetzungsprofile, sowie ihre Biokompatibilität waren sehr vielversprechend. In den zugehörigen in-vitro-Studien konnte ein Chemotherapeutikum erfolgreich in Krebszellen transportiert werden.
