Abstract:
Pore-forming toxins (PFT) are virulence factors that transform from soluble to membrane-bound states. The Yersinia enterocolitica YaxAB system represents a family of binary α-helical PFTs with orthologues in human, insect and plant pathogens, with so far unknown structures. The goal of this thesis was the thorough structural and biochemical characterization of the YaxAB toxin. To achieve this, a multi-disciplinary approach was taken, combining different structural methodologies to reveal architectures of both the soluble components as well as the reconstituted pore complex.

Porenbildende Toxine sind Virulenzfaktoren welche von löslicher zu membrangebundener Form transformieren können. Das YaxAB System von Yersinia enterocolitica stellt eine Familie zweiteiliger Porentoxine dar, mit Orthologen in Krankheitserreger von Menschen, Insekten und Pflanzen. Strukturen von YaxAB verwandter Toxine sind bisher jedoch nicht bekannt. Das Ziel dieser Doktorarbeit war eine detaillierte strukturelle und biochemische Untersuchung des YaxAB Toxins. Hierfür wurde ein multidisziplinärer Ansatz verwendet der aus einer Kombination verschiedener struktureller Methoden bestand, um sowohl die Struktur der einzelnen Toxinuntereinheiten, wie auch die der rekonstituierte Toxinpore, zu ermitteln.
