Cobalamins are vitamers of vitamin B12. In the present thesis, a biosynthetic approach was developed for the preparation of 15N-labelled cobalamins employing Propionibacterium freudenreichii. Using the biosynthetically prepared 15N-labelled cobalamins as internal standards, the first multiple stable isotope dilution assay (SIDA) was developed to simultaneously determine four naturally occurring cobalamins in meats. The last part of the thesis compared the novel method analyzing the native cobalamins and a conversion SIDA method measuring total B12 after cyanidation.
