Abstract:
HECT E3 ligases play a crucial role in regulating cellular processes through linkage-specific ubiquitin chain formation. Malfunctions contribute to various diseases, yet the structural mechanisms remain elusive.
Employing chemical probes, and cryo-EM, we elucidated a detailed HECT polyubiquitylation cascade. This study reveals mechanistic intricacies of UBR5 showcasing its robust catalytic activity, remarkable processivity, and sustained linkage-specificity during ubiquitin chain formation.

HECT-E3-Ligasen spielen eine entscheidende Rolle bei der Regulierung zellulärer Prozesse durch die verknüpfungsspezifische Bildung von Ubiquitin-Ketten. Fehlfunktionen tragen zu verschiedenen Krankheiten bei, doch die strukturellen Mechanismen sind nach wie vor schwer zu verstehen.
Mit Hilfe von chemischen Sonden und Kryo-EM haben wir eine detaillierte HECT-Polyubiquitinylierungskaskade aufgeklärt. Diese Studie enthüllt mechanistische Feinheiten von UBR5, die seine robuste katalytische Aktivität, bemerkenswerte Prozessivität und anhaltende Verknüpfungsspezifität während der Ubiquitin-Kettenbildung aufzeigen.
