Schwerpunkte der Arbeit sind einerseits Strukturanalyse und Dynamik, andererseits die nähere Untersuchung der in den letzten Jahren neu entwickelten Methode der dipolaren Kopplungen. Im Rahmen der Dissertation wurde die aktive Domäne des Herpes Simplex Virus Proteins ICP47 mittles NMR aufgeklärt. Sie besteht in der Umgebung von Membranen aus zwei alpha-Helices, die durch einen ungeordneten Bereich miteinander verbunden sind. Ein weiteres Kapitel beschäftigt sich mit der Dynamik des Proteinrückgrats des Blutegel Carboxypeptidasen Inhibitors LCI. Er weist sowohl ungebunden als auch gebunden an die menschliche Carboxypeptidase A2 eine hauptsächlich starre Struktur auf. Im letzten Kapitel werden Theorie und experimentelle Methoden für die Bestimmung und Anwendung der residualen dipolaren Kopplungen beschrieben.

Three main topics of NMR were investigated: Structure and dynamic of two proteins on the one side and the new method of residual dipolar couplings on the other. The first chapter describes the structuredetermination of the active domain of the Herpes Simplex Virus protein ICP47. ICP47 consists in a membranelike environment of two alpha-helices which are connected by an unoriented loop. The backbonedynamic of LCI (Leech Carboxypeptidase Inhibitor) is investigated in the second part. Free and in complex with the human carboxypeptidase A2 it shows a very rigid structure. Theory and experimental methods for determination and application of the residual dipolar couplings are described in the last chapter.
