The role of chicken αβ and γδ T cells in the defense against infection is largely unknown. Reverse genetics can be used to study the functions of individual lymphocyte populations. The phenotype of TCR Cβ KO chickens showed an imbalance of humoral and cytotoxic immune defense. Furthermore, using TCR Cγ KO chickens during infection with Eimeria tenella, it was shown that TCR2+ αβ T cells can compensate for the loss of γδ T cells. mCherry+ T cells were tested for adoptive transfer of different cell numbers and tissues. In addition, Cre-ERT2 chimeras and a loxP TCR Cβ Knockin germ cell line were generated to create an inducible TCR Cβ KO system.
