Abstract:
Der humane Transkriptionsfaktor Forkhead Box Protein P2 (FOXP2) spielt in der Entwicklung menschlicher Sprache eine bedeutende Rolle. Durch eine Mutation (R553H) in seiner Forkhead-Domäne wird die Funktionsfähigkeit des Transkriptionsfaktors zerstört und eine genetisch bedingte Sprach- und Sprechstörung ausgelöst. Ziel dieser Arbeit war die Untersuchung der Motive und Domänen von FOXP2 ohne den störenden Einfluss zellulärer Bindungspartner. Mit Hilfe der biochipbasierten switchSENSE-Technologie wurden zeitaufgelöste Interaktionsanalysen durchgeführt, um die DNA-Bindung hinsichtlich der geleisteten Beiträge der Motive und Domänen zu charakterisieren.

The human transcription factor forkhead box protein P2 (FOXP2) plays an important role in the development of human language and speech. A mutation (R553H) in its forkhead domain destroys the function of the transcription factor and triggers a genetic speech and language disorder. The aim of this work was to investigate the motifs and domains of FOXP2 regarding their DNA-binding ability without interfering influences of cellular binding partners. In order to characterize DNA-binding in respect of the contribution of motifs and domains, time-resolved interaction analyses were performed using the biochip-based switchSENSE technology.
