Abstract:
The 26S proteasome is a 2.5 MDa protein complex that orchestrates protein degradation in eukaryotic cells at the final stage of the ubiquitin proteasome system (UPS). A combination of different approaches – provision of nucleotide analogs, directed point mutations in the AAA+ ATPase subunits or using a designed model substrate – resulted in the alteration of the conformational equilibria of the proteasome, and ultimately with the help of single-particle cryo-electron microscopy led to the identification of three newly discovered proteasome states. With this the communication between the proteasome subunits could be better understood.

Das 26S Proteasom ist ein 2,5 MDa großer Proteinkomplex, der als Exekutive im Ubiquitin-Proteasom-System (UPS) für den Abbau von Proteinen in eukaryotischen Zellen verantwortlich ist. Die Kombination dreier Methoden – der Einsatz von Nukleotidanalogen, direkte Punktmutationen im ATPase Modul und die Zugabe eines Modell-Substrats – wurde benutzt, um das 26S Proteasom in unterschiedliche Zustandsequilibria zu überführen und schließlich mit Einzelpartikel-Kryo-Elektronenmikroskopie drei unbekannte Zustände zu entdecken. Mit Hilfe dieser konnte die Kommunikation zwischen den 26S Proteasom-Untereinheiten besser verstanden werden.
