RNAi-mediated suppression of PD-L1 to restore Hepatitis B Virus-specific immunity and enhance the efficacy of therapeutic vaccination

Chronically HBV-infected patients are at risk for liver cirrhosis and hepatocellular carcinoma. This thesis shows a promising approach to eradicate HBV by inhibiting the immunosuppressive protein PD-L1 in combination with therapeutic vaccination. In contrast to previous work, a siRNA-based approach was used, which allowed an organ-specific immunomodulation as well as an enhanced therapeutic efficacy compared to antibody mediated checkpoint inhibition.
