We studied the role of active zone proteins in the synaptic vesicle distribution and presynaptic cytomatrix organization by using cryo-Electron Tomography. Automated detection and comparative quantitative analysis of connectors, tethers and vesicles showed that RIM protein is required for bringing the vesicles to the region proximal to the AZ membrane, while Munc13 and SNAP25 proteins tether and localize vesicles at distances closer than 10nm and 5nm from the AZ membrane, respectively.
