Abstract:
Ephrin-A5 ist ein membranständiges 25 kDa-Glykoprotein, das einen Liganden der Eph-Subklasse von Tyrosin-Kinase-Rezeptoren darstellt. In dieser Arbeit wurden Experimente zur Expression und Funktion dieses Moleküls in der Maus durchgeführt. RNA-in situ-Hybridisierungen zeigen, daß Ephrin-A5 u.a. im cerebralen Cortex, Hypothalamus, Cerebellum und olfaktorischen System exprimiert wird. Die phänotypische Analyse Ephrin-A5-defizienter Mäuse, die über homologe Rekombination in ES-Zellen hergestellt wurden, lieferte Hinweise auf die Funktion von Ephrin-A5 in diesen Organstrukturen. So treten bei einem Teil der homozygoten Ephrin-A5-Mutanten schwere Neuralrohrdefekte auf. Ausserdem konnten Faltungsdefekte des Cerebellums und Veränderungen in den thalamocorticalen Projektionen festgestellt werden.

Ephrin-A5 is a membrane associated 25 kDa glycoprotein that serves as a ligand for receptor tyrosine kinases of the Eph subclass. In this work experiments have been performed to reveal expression and function of this molecule in the mouse. RNA in situ hybridization has shown that Ephrin-A5 is expressed among other domains in the cerebral cortex, the hypothalamus, the cerebellum, and the olfactory system. The phenotypic analysis of ephrin-A5 deficient mice, that where generated by homologous recombination in ES cells, gave information on the function of Ephrin-A5 in these structures. For instance a minority of homozygous ephrin-A5 mutants displays severe neural tube defects. In addition foliation defects in the cerebellum and alterations in the thalamocortical projections have been observed.
