Abstract:
The so far unexplored, cytotoxic natural product neocarzilin A (NCA) was detected to inhibit cell proliferation and cell migration of human breast cancer cells. ABPP (Activity-based protein profiling) approaches identified VAT-1 as cellular target protein of NCA. Orthogonal approaches confirmed the identity of the protein and detected the region involved in natural product binding. Further, VAT-1, which is mainly relevant for inhibition of cell migration, was revealed to be intricated in a network of proteins involved in cell adhesion by mass spectrometry-based co-immunoprecipitation.

Eine Hemmung der Zellproliferation und Migration von humanen Brustkrebszellen wurde für den bisher unerforschten, cytotoxischen Naturstoff Neocarzilin A (NCA) gezeigt. VAT-1 wurde als zelluläres Zielprotein von NCA durch ABPP (Aktivitäts-basiertes Protein-Profiling) identifiziert. Orthogonale Ansätze bestätigten das Zielprotein und die Region in der Proteinsequenz, die für die Interaktion relevant ist. Zusätzlich zeigte eine Massenspektrometrie-basierte Ko-Immunopräzipitation, dass VAT-1, welches vor allem für die Hemmung der Zellmigration verantwortlich ist, in einem Netzwerk von Zelladhäsions-Proteinen verwickelt ist.
