Abstract:
Staphylococcus aureus is a major bacterial pathogen. AV73 did not only reduce alpha-hemolysin production in S. aureus, but also impeded in vitro biofilm formation. Bacterial proteomes and extracellular protein levels were analyzed and possible targets identified via affinity-based protein profiling. Caseinolytic protease P (ClpP) plays a major role in S. aureus virulence. While inhibition of ClpP leads to a decrease in virulence, activation has a bactericidal effect. Staphylococcus aureus ist ein wichtiger bakterieller Krankheitserreger. AV73 reduziert nicht nur die Bildung von alpha-Hämolysin in S. aureus, sondern inhibiert auch die Biofilmbildung in vitro. Mögliche Zielproteine wurden mittels affinitäts-basiertem Protein-Profiling analysiert. Caseinolytische Protease P (ClpP) spielt eine wichtige Rolle in der Virulenz von S. aurues. Während ihre Inhibition zu einem Rückgang in der Viurulenz führt, hat ihre Aktivierung eine bakterientötende Wirkung. Beide Themen wurden in dieser Arbeit behandelt.
