Abstract:
To visualize global molecular mechanisms that ensure a successful immune response, a comprehensive kinetic study determining changes in RNA polymerase II (RNAPII) binding (ChIP-seq), transcription and splicing (RNA-/4sU-seq), and translation (ribosome profiling) was performed during activation of in vitro generated Th1 cells. Rapid de novo recruitment of RNAPII primarily mediates transcriptional changes, which are coupled to translation. Cotranscriptional splicing rates show a genome-wide temporary slowdown, that is subsequently optimized and correlates with increasing Ser2 phosphorylation of the RNAPII CTD and activation of the positive transcription elongation factor (pTEFb).

Um globale molekulare Mechanismen, die eine erfolgreiche Immunantwort garantieren, zu untersuchen, wurde eine umfassende kinetische Studie zur Bestimmung von Veränderungen der RNA-Polymerase-II-(RNAPII)-Bindung (ChIP-seq), Transkription und Spleißen (RNA-/4sU-seq) und Translation (ribosome profiling) während der Aktivierung von in vitro generierten Th1-Zellen durchgeführt. Eine schnelle Neurekrutierung der RNAPII ist für Änderungen der Transkription verantwortlich, die mit Translation gekoppelt sind. Das kotranskriptionelle Spleißen ist genomweit vorübergehende verlangsamt, was anschließend wieder optimiert wird und mit zunehmender Ser2 Phosphorylierung der RNAPII CTD und der Aktivierung des positiven Transkriptionselongationsfaktors (pTEFb) korreliert.
