Das Hepatitis-B-Virus (HBV) besitzt eine strikte Speziesspezifität. Die Ergebnisse dieser Arbeit zeigen, dass das Na+-Taurocholat cotransportierende Polypeptid (NTCP) der limitierende Faktor einer HBV-Infektion von Makaken- und Schweine-Hepatozyten ist. Mittels eines neu etablierten, quantifizierbaren Expressions-Systems konnten darüber hinaus NTCP-Varianten verschiedener Tierspezies als funktionelle HBV-Rezeptoren identifiziert werden. Diese Arbeit liefert somit wichtige Erkenntnisse zum Verständnis der HBV-Interspezies-Transmission und ermöglicht die Etablierung neuer HBV-Infektionsmodelle.     «

Hepatitis B virus (HBV) possesses strict species specificity. This thesis shows that the Na+-taurocholate cotransporting polypeptide (NTCP) is the limiting factor for HBV infection in macaque and porcine hepatocytes. Moreover, using a newly established quantitative expression system, it was possible to compare NTCP variants and to identify NTCPs of different species as functional HBV receptors. Taken together this thesis enables new insight into the inter-species transmission of HBV and paves the way for the establishment of new HBV animal models.     «
