Abstract:
Non-invasive imaging and manipulation techniques are important tools to understand causal relationships in cellular networks. To visualize genetically defined cells via magnetic resonance imaging, I genetically modified the transferrin, the ferrous iron, and the ferritin uptake pathway and evaluated expressed ferritin as cellular iron storage. The selective uptake of magnetoferritin also allowed for magnetomechanical manipulation of cells as was shown by magnetic cell sorting and cell ablation via magnetic hyperthermia.

Nicht-invasive Bildgebungs- und Manipulationstechniken sind wichtige Werkzeuge, um kausale Beziehungen in zellulären Netzwerken zu verstehen. Um genetisch definierte Zellen anhand von Magnetresonanzbildgebung zu visualisieren, wurden die Tranferrin-, die Eisen(II)- und die Ferritinaufnahme genetisch verändert und exprimiertes Ferritin als zellulärer Eisenspeicher evaluiert. Die selektive Aufnahme von Magnetoferritin ermöglichte die magnetomechanische Manipulation von Zellen, wie durch magnetische Zellsortierung und magnetische Hyperthermie induzierte Zellabtötung gezeigt wurde.
