The advent of immunotherapy in the treatment of cancer shed new light on the importance of modulating the immune system in order to achieve a therapeutic response, in addition to traditional treatments targeting the tumor. This thesis focuses on the study of main players of anti-cancer response, T-cells, and on the identification of their ideal molecular targets on the tumor surface, neoantigens. Acquired knowledge on functional and molecular features of T cells and respective neoantigens will contribute to the development of novel personalized T-cell-based immunotherapies.

Das Aufkommen der Immuntherapie bei der Behandlung von Krebs wirft ein neues Licht auf die Bedeutung der Modulation des Immunsystems zur Erzielung einer therapeutischen Reaktion. Immuntherapie wirkt zusammen mit herkömmlichen Behandlungen, die auf den Tumor direkt abzielen. Diese Dissertation konzentriert sich auf die Untersuchung der Hauptakteure der Anti-Krebs-Reaktion, T-Zellen, und auf die Identifizierung ihrer idealen molekularen Ziele auf der Tumoroberfläche, Neoantigene. Das erworbene Wissen über funktionelle und molekulare Eigenschaften von T-Zellen und entsprechenden Neoantigenen wird zur Entwicklung neuartiger personalisierter T-Zell-basierter Immuntherapien beitragen.
