Kurzfassung:
Oncolytic virus therapy and adoptive T-cell therapy have both recently shown clinical success in cancer patients. We hypothesized that a combination of both approaches, in particular, the use of T-cell receptor (TCR) transgenic T-cells as cellular carriers to chaperone oncolytic vesicular stomatitis virus (VSV) would not only shield the virus from inactivation and provide tumor specificity in virus delivery and transfer, but the carrier cells would exert a direct antitumor effector function themselves. This work provides preliminary in vitro data in support of this approach.

Onkolytische Virotherapie und adoptiver T-Zell-Transfer finden erfolgreich Anwendung in der modernen Tumortherapie. Dennoch gibt es Hürden, die wir durch eine Kombination beider Therapieansätze überwinden möchten. Dafür kommen T-Zell-Rezeptor transgene T-Zellen als zelluläre Transporter zum Einsatz, welche die onkolytischen Viren im Blutstrom vor inaktivierenden Komponenten schützen. Gleichzeitig ermöglichen sie eine zielgerichtete Beförderung zum Tumor, um ihn dort gemeinsam zu bekämpfen. Diese Arbeit stellt erste Daten bereit, um dieses Konzept zu unterstützen.
