After optimization and standardization of 2D-electrophoresis with immobilized pH gradients (IPG-Dalt), the proteome analysis of three wild-type strains of yeast, as well as a reproducibility study with the strain FY1679 was conducted by the AG Proteomik with the involvement of two other laboratories. After finalization of the methodology, 28 deletion mutants of yeast, as well as the cold stress reaction of two S. cervisiae strains were analyzed to characterize the ORFs of unknown function. One of the focal points of this study was the optimization of the 2D-electrophoresis methodology for the analysis of the S.cervisiae proteome. This involved the development of narrow, immobilized pH gradients (IPGs), as well as the first time discussed reproducibility of separating strong alkaline yeast proteins. Over a 100 of these proteins could be identified using MALDI-TOF-MS and an internet 2D-database was established.
