Monoclonal antibodies were developed against domoic acid (DA). DA is an algal toxin which is produced by the diatom Pseudonitzschia pungens f. multiseries. It is accumulated by blue mussels and upon ingestion, it causes the human illness syndrome Amnesic Shellfish Poisoning (ASP), which is characterized by neurological syndromes, such as headache, confusion, disorientation and deficits in short-term memory. For the detection of domoic acid poly- (pab) and monoclonal antibodies (mab)were developed. Enzym-linked immunoassays (ELISA)were carried out with the pab and the mab. Detection limits of 0,8 and 4 µg/l were obtained,respectively. No crossreactivities against similar chemical structures and algal toxins could be detected for the pab and the mab. Moreover these test formats were well suited to detect DA in spiked mussel extracts. Mass production of mab was carried out with fermenter systems. The antigen-antibody interaction of the mab were carried out with the BIAcore. Furthermore a competitive immunoassay was developed with the BIAcore. A detection limit of 1,2 µg/l could be obtained with this test format.
