Action potentials propagate in axons of neurons but they also backpropagate into the dendrites. In this work those backpropagating action potentials were measured in cultured hippocampal neurons with voltage-sensitive dyes. They were optically recorded up to 400µm from the soma at a resolution of 5µm (90*60 pixel) and 0,75ms without averaging. It was shown, that trains of backpropagating action potentials exhibit no activity-dependent attenuation of the spike amplitude as in brain slices but a broadening of the spike width. Additionally, dendritic action potentials can be altered by transfecting the neurons with voltage gated ion channels (Kv1.3, Kv1.4).
