Inform einer klinisch-experimentellen Studie wurde an 50 Patienten, die sich einer Koronarstentimplatntation unterzogen, die Wirksamkeit der antithrombozytaeren Substanzen Ticlopidin (Dosis: 2x500mg initial, dann 2x250mg taeglich) und Clopidogrel (75mg taeglich ohne bzw. mit 300, 450 oder 600mg Ladungsdosis) verglichen. Das Patientenblut wurde vor Einnahme sowie zwei, vier, acht, 24 und 48 Stunden spaeter in der Aggregometrie nach Born sowie durchflusszytometrisch auf die thrombozytaere Aktivitaet ueberprueft. Beide Messverfahren zeigten, dass Clopidogrel – insbesondere mit den Ladungsdosen 450 und 600 mg – eine im Vergleich zu Ticlopidin sowie Clopidogrel ohne Ladungsdosis signifikant verminderte Aggregierbarkeit der Blutplaettchen sowie Expression aktivierter thrombozytaerer Fibrinogenrezeptoren (PAC-1) bewirkt.     «

A clinical-experimental study was performed in order to compare in 50 patients undergoing coronary stent placement the effectiveness of the antiplatelet substances Ticlopidine (dose: 2x500mg initially, then 2x250mg daily) and Clopidogrel (75mg daily without and with 300, 450 or 600mg loading dose). Blood samples were analysed on platelet activity before as well as two, four, eight, 24 and 48 hours after administration by Born´s aggregometry and by flowcytometry. Both procedures showed that Clopidogrel – especially with the loading doses of 450 and 600 mg – reveales an accelerated effect of significantly diminished platelet aggregation compared to Ticlopidine as well as Clopidogrel without loading dose as well as expression of activated platelet fibrinogen receptors (PAC-1).     «
