Zur Etablierung von Primärzellkulturen wurden Zellen aus Aszites und Pleuraergüssen von 24 Patienten mit fortgeschrittenen gastrointestinalen Tumoren gewonnen, 19 Zelllinen wurden kultiviert und charakterisiert. Die Tumorzellen wurden durch Verwendung magnetgekoppelter Antikörper bzw. Klonierungsverfahren separiert. Der Tumorzellnachweis wurde mittels DNA-Analyse bzw. durchflusszytometrischer Messung von Zellmembranantigenen geführt. Der niedrige Differenzierungsgrad konnte teilweise durch den Einsatz von Differenzierungsinduktoren erhöht werden. Die Hormone Bombesin und Somatostatin wurden im Kulturmediumüberstand gemessen, die p53-Proteine in Tumorzellen dargestellt.     «

In order to set up primary cell cultures cells were taken from ascites and pleura effusions of 24 patients suffering from advanced gastrointestinal tumors, out of which 19 cell lines were cultivated and characterised. The tumor cells were separated by using magnet bounded antibodies and the according cloning procedures. The tumor cells were proved with the help of DNA-analysis respectively by measuring the antigenes of the cell membranes. The low grade of differenciation could partly be raised by means of differenciation inductors. The hormons Bombesin and Somatostatin were measured in overmature cultures, the p53-proteines were described in tumor cells.     «
