Subject of this thesis was the development, optimization and preclinical evaluation of novel protein-based reagents for tumor diagnostic via PET. An aldehyde-chemistry based site-specific coupling procedure was established for the radiolabeling of Anticalins. The target affinity of two ED-B specific Anticalins was improved by factors of 120 and 230 via protein engineering. Optimization of the plasma half-life of Fab fragments directed against the tumor antigens HER2 and CD20 via PASylation resulted in significant improvement of tumor-uptake. Hence, Fab-PAS200 presents a promising tracer format for clinical translation.
