PAS-polypeptides with different sequences of Pro, Ala and/or Ser were prepared by recombinant DNA technology both in free form – by means of cleavable fusion proteins – as well as fused to the therapeutic protein IL-1Ra with the goal of application as biomimetics for poly-ethylene glycol (PEG). Characterization of hydrodynamic, conformational and hydrophilic properties as well as polymer viscosity indicated surprising similarity between these biopolymers and PEG, which opens interesting perspectives for biopharmaceutical use. Furthermore, in the case of special PAS-polypeptides applicability for chemical conjugation with small molecules and as an MS calibration standard were demonstrated.
