Für den Nachweis von 38 Polymorphismen in 17 Kandidatengenen wurden Real-Time-PCR-Methoden entwickelt und 585 Psychiatrie-Patienten genotypisiert. Mittels HPLC-MS/MS wurden die Serumkonzentrationen der sechs häufigsten Psychopharmaka bestimmt. Für CYP450-Gen-Polymorphismen zeigten sich Unterschiede in den Serumwerten, was sich zum Teil in Assoziationen zu Response- und Nebenwirkungsunterschieden widerspiegelte (CYP2D6 und CYP2C19). Bestimmte Serotonintransporter-Genvarianten waren bei SSRI-Patienten mit mehr Nebenwirkungen, geringeren Ansprechraten und längeren Behandlungszeiten assoziiert.     «

Real-time PCR methods for the detection of 38 polymorphisms in 17 candidate genes have been established and 585 psychiatric inpatients were genotyped. Serum concentrations of the 6 most commonly used psychotropics were analysed by HPLC-MS/MS. CYP450 gene polymorphisms were associated with different serum concentrations and partly, differences in response and side effects were identified (CYP2D6 and CYP2C19). In pharmacodynamic structures, notably serotonin transporter variants were linked with higher side effect rates, reduced response and prolonged hospitalization in patients with SSRI.     «
