This thesis shows that the post-translational protein modification Nedd8 is active in the brain and that PSD-95, the main scaffold of the post-synaptic density, is directly neddylated. A newly developed mass spectrometry approach identified the neddylated lysine residues of PSD-95 and electrophysiological studies revealed that PSD-95 neddylation is required for AMPA receptor mediated neurotransmission. In conclusion, this thesis demonstrates that neddylation is an important post-translational modification in the brain controlling synaptic function.
