Adult neurogenesis occurs in the subventricular zone (SVZ) of the lateral ventricles. In this neurogenic niche new interneurons of the olfactory bulb (OB) are generated. In this project, the molecular mechanisms regulating the generation of dopaminergic periglomerular interneurons were investigated. In addition, the signalling pathways controlling the integration of new neurons into the olfactory bulb neural network were analyzed. Gain-of-function experiments in vitro and in vivo showed, that Wnt1-protein increases proliferation of neuroblasts in the SVZ-OB-system via the canonical Wnt-pathway. Also the generation of new dopaminergic periglomerular interneurons was increased in response to overexpression of Wnt1. Furthermore, it was determined that the CREB-signal transduction pathway in vivo controls the expression of neuronal differentiation genes like Doublecortin and Pax6 and that the CREB-pathway controls the integration of newborn neurons into the pre-existing neural network. Hence, the Wnt and the CREB signalling pathways both contribute to the control of adult neurogenesis in the SVZ-OB-system.
