The results presented in this thesis describe a new role for STAT3 (signal transducer and activator of transcription 3) as an integral component of TLR mediated signal transduction. STAT3 interacted with the cytoplasmic TIR domain of TLRs as well as with the adapter molecule MyD88. TLR induced activation of mTOR (mammalian Target of Rapamycin ) led to STAT3 serin 727 phosphorylation and resulted in dissociation of STAT3 from its complex with MyD88. Upon serin phosphorylation, STAT3 translocated into the nucleus and induced, upon promoter binding, transcription of PAI-2 (plasminogen activator inhibitor 2).
PAI-2 protein interacted with ASC and thus inhibited the formation of NALP3 and AIM2 dependent inflammasomes, thus dampening the release of bioactive IL-1b.
