The Notch pathway plays an important role in pancreatic development, during exocrine regeneration as well as KrasG12D-driven pancreatic ductal adenocarcinoma development. To analyze the role of HES1, a key downstream target of the Notch signaling pathway, a genetically engineered mouse model with a pancreas-specific deletion of Hes1 was generated. A key role of HES1 in acinar cell integrity and plasticity during cell maturation and regenerative processes after acute pancreatitis could be demonstrated. In KrasG12D-driven carcinogenesis HES1 appears to promote a PanIN to PDAC route.
