The aim of this work was to increase the efficiency of in vitro gene transfer for eukaryotic cells. To this end, the effect of all steps during the transfection process on the transfection efficiency had to be evaluated. Multiple critical steps were identified, which influence strongly the transgene expression to a different degree depending on the type of cells used.
Those steps were the binding of transfection complexes to the cells, the internalization of transfection complexes by the cells and the transport of DNA into the nucleus.
All limiting steps were optimized through the use of adequate peptides, which lead to an improved transfection efficiency due to synergistic effects.
