The aim of this study was to identify genes regulated by IFN gamma and TNF that play a role in the immune defense against intracellular microbes. Using microarray analyses, expression profiles of murine macrophages stimulated with these cytokines were analyzed. Thereby, IRG1 and mGBP6 were identified as highly induced genes. Both gene transcripts are highly upregulated in vitro under proinflammatory conditions, and in vivo after infection with intracellular pathogens. By using confocal microscopy, IRG1 was identified as mitochondria-associated protein, whereas mGBP6 is localized in cytoplasmic vesicles. After in vitro infection of mGBP6 expressing macrophages with Toxoplasma gondii, mGBP6 associates with the parasitophorous vacuole of the parasite, suggesting a direct Interaction of the protein with the pathogen.
