The use of livestock animals to model human cancers depends on the ability to control expression of mutated oncogenes or tumour suppressor genes in a spatial- and temporal-specific manner. In this work, different approaches to create pancreas-,lung- and gut-specific Cre-driver pig lines for cancer research were investigated, using PDX-1, SFTP-C and villin-1 promoters, respectively. Also, aiming at controlling the Cre activity in a temporal manner, 2 tamoxifen-inducible forms of Cre were compared in vitro.

Um menschliche Krebserkrankungen in Großtieren akkurat modellieren zu können, ist eine zeitlich und räumlich spezifische Gendeletion oder Expression von fundamentalem Nutzen. In dieser Arbeit wurden verschiedene Ansätze durchgeführt und analysiert um Pankreas-,Lungen- & Kolongewebe-spezifische Cre-Rekombinase-Expression in Schweinen für die Krebsmodellierung zu generieren. Um die Cre-Aktivität in einer zeitlichen Weise zu steuern, 2 Tamoxifen-induzierbare Formen der Cre wurden in vitro verglichen.
