Abstract:
Inflammatory bowel diseases (IBD) are chronic intestinal inflammations with constantly rising incidences that are associated with Western lifestyle. The two major phenotypes are Crohn’s disease and ulcerative colitis. In the present study, the influence of CCL17 on the development of experimental colitis in mice, resembling human disease, was assessed using CCL17 knockout mice. Thus, it was shown that CCL17 plays a disease promoting role, which could be demonstrated on histological, cell biological and molecular level. Taken together, this study indicates that neutralizing CCL17 might be a promising target for future treatment of IBD.

Chronisch entzündliche Darmerkrankungen (CED) treten in stetig wachsender Häufigkeit auf und sind mit dem westlichen Lebensstil assoziiert. Die zwei häufigsten Ausprägungen der Erkrankung sind Morbus Crohn und Colitis Ulcerosa. In der vorliegenden Arbeit wurde der Einfluss von CCL17 auf die Entstehung experimenteller Colitis in Mäusen, mit Hilfe von CCL17-knock-out Mäusen, untersucht. Hierbei konnte gezeigt werden, dass CCL17 einen entzündungsfördernden Einfluss auf den Krankheitsverlauf hat. Dies wurde mittels histologischer, zellbiologischer und molekularbiologischer Methoden bewiesen. Abschließend bleibt festzuhalten, dass die Neutralisierung von CCL17 einen neuen potentiellen Ansatz für die Therapie von CED darstellt.
