Abstract:
Capillary electrophoresis (CE) coupled to mass spectrometry (MS) is becoming increasingly important as analytical separation technique. In this thesis, 2D separation methods were developed in order to overcome limitations of CE-MS such as potential interference with electrospray ionization, low concentration sensitivity, and in particular cases deficient resolution and separation efficiency. These 2D methods include CE-CE-MS, liquid chromatography (LC)-CE-MS and CE-ion mobility mass spectrometry (IM-MS) for the analysis of small pharmaceuticals, intact proteins and glycans.

In dieser Thesis wurden 2D Trennmethoden entwickelt um mit CE-MS einhergehende Einschränkungen wie eine potenzielle Störung der Elektrospray Ionisierung, geringe Konzentrationssensitivität und in besonderen Fällen unzulängliche Auflösung und Trenneffizienz zu überwinden. Diese 2D Methoden beinhalten CE-CE-MS, Flüssigkeitschromatographie (LC)-CE-MS und CE-Ionenmobilitäts-Massenspektrometrie (IM-MS) für die Analyse von kleinen pharmazeutischen Molekülen, intakten Proteinen und Glykanen.
