Pigs carrying an
APC mutation orthologous to the human
APC1309 mutation frequently diagnosed in human colorectal cancer were characterised by mRNA and miRNA sequencing to identify modifier genes of severe polyposis and genes driving tumour progression. The gene
CYP7A1, miRNAs miR-215 and 194b-5p were associated with severe polyposis and the genes
PLXND1 and
GBP6 and miRNAs let-7e, miR-146a-5p, 146b, 183, 196a with tumour progression. The
CAS9 gene was placed at the
ROSA26 locus of
APC1311 cells to generate cloned piglets, in which gene editing can then be carried out directly in the polyps to accelerate tumour progression.
