Microbes capable of breaking down hydrocarbon contaminants are widespread in nature, but the contributions and functions of those employing microaerobic or unusual metabolic or respiratory strategies are not well understood. In this thesis, I investigated the potential for oxygenesis and oxygen-activation, nitrogen respiration (OANR) in toluene degradation. To do this, I developed a novel method of semi-quantitative total-RNA-stable isotope probing (SIP) that improves upon traditional SIP by providing data on both identity and function of process-relevant microbes.

Kohlenwasserstoff-abbauende Mikroorganismen sind in der Natur weit verbreitet. Allerdings ist die Bedeutung von Abbauern mit besonderen Stoffwechselanpassungen in natürlichen Systemen oft noch wenig verstanden. Diese Doktorarbeit untersucht zwei dieser Anpassungen, die postulierte Oxygenese, sowie den Sauerstoff-aktivierenden, Nitrat-atmenden Abbau. Zudem wird eine neue Methode zur semi-quantitativen Analyse von 13C-markierten Transkriptomen entwickelt, welche sowohl die Identität als auch die Genexpression prozessrelevanter Mikroben in natürlichen Systemen abbildet.
