Abstract:
Pheochromocytomas (PCCs) and paragangliomas (PGLs), commonly referred to as PPGLs, as well as pancreatic neuroendocrine tumors (pNETs), belong to the group of neuroendocrine tumors. We found dysregulation of PI3K/AKT and the CDK4/6 pathway in these tumors. Consequently, we could show that the treatment of both PPGLs and pNETs with PI3K and CDK4/6 inhibitors is highly effective and has clinical potential. Additionally, we could characterize PPGL tumors occurring in MENX rats and show that PGLs show a more aggressive behavior than PCCs.

Phäochromozytome (PCCs) und Paragangliome (PGLs) sowie pankreatisch neuroendokrine Tumore (pNETs) gehören zur Gruppe der neuroendokrinen Tumore. Wir konnten eine Fehlregulation des PI3K/AKT sowie des CDK4/6 Signalweges in diesen Tumoren beobachten und zeigen, dass ein Behandlungsansatz mit Inhibitoren dieser Signalwege sehr effektiv ist und klinisches Potential aufweist. Zusätzlich konnten wir auch PPGLs, die sich in MENX Ratten entwickeln, weitergehend charakterisieren. Unsere Ergebnisse haben gezeigt, dass PGLs ein aggressiveres Verhalten als PCCs aufweisen.
