MLL rearrangements are frequently detected in childhood acute lymphoblastic leukemia (ALL) leading to an aggressive type of leukemia with bad prognosis. In this study, I investigated the role of the two chimeric fusion proteins, MLL/AF4 and AF4/MLL, for leukemia maintenance using breakpoint specific shRNAs in patient-derived xenograft acute leukemia cells. In vivo experiments with a constitutive and an inducible Cre-ERT2 based knockdown system revealed an essential function of MLL/AF4 in tumor maintenance. Thus, targeting MLL/AF4 using RNAi might be a specific and promising therapeutic solution improving prognosis and outcome of patients suffering from t(4; 11) ALL.
