Multiple Sclerosis (MS) is an autoimmune disease of the brain and spinal cord. In healthy conditions, the infiltration of immune cells into the central nervous system (CNS) is controlled by the blood-brain barrier (BBB). In MS, however, activated T cells can cross the BBB, recruit and instruct monocyte-derived macrophages that contribute to tissue damage. In my thesis, I used CRISPR gene editing in MS models to perform a comprehensive characterization of two critical steps in the disease pathogenesis, the CNS infiltration of T cells and the local instruction of phagocyte phenotypes.

Multiple Sklerose (MS) ist eine Autoimmunerkrankung des Gehirns und des Rückenmarks. Unter gesunden Bedingungen wird das Eindringen von Immunzellen in das Zentralnervensystem (ZNS) durch die Blut-Hirn-Schranke (BBB) gesteuert. Bei MS jedoch können aktivierte T-Zellen die BHS passieren, aus Monozyten stammende Makrophagen rekrutieren und anweisen, die zu Gewebeschäden beitragen. In meiner Doktorarbeit habe ich die CRISPR-Geneditierung in MS-Modellen verwendet, um eine umfassende Charakterisierung von zwei kritischen Schritten in der Krankheitsentstehung durchzuführen, der ZNS-Infiltration von T-Zellen und der lokalen Instruktion von Phagozyten-Phänotypen.
