The aim of this work was the selection, characterization and partial optimization of suitable enzymes to design an
in-vitro production system for industrially relevant solvents, such as isobutanol and
n-butanol, on the basis of renewable resources. To accomplish this target, each enzyme component was examined in regard to its catalytic properties, process stability, and cofactor requirements. Particularly, the enzyme activities of ketol-acid reductoisomerase, thiolase and crotonase were in the focus.
