The molecular chaperone BiP assists in the folding and assembly of secreted proteins like antibodies and is regulated by several cofactors such as the nucleotide exchange factor BAP (Sil1). This study analyzed its effects on the conformations and the catalytic cycle of BiP. Moreover, the mode of substrate recognition by BiP was investigated and novel binding sites in new substrates were identified and their binding affinities determined. This allows a better understanding of how BiP interacts with cofactors and substrates.
